The formulation contains selective agents such as crystal violet and thallium sulfate that suppress contaminating organisms commonly present in raw milk, allowing streptococci to grow more clearly. Differentiation is primarily based on esculin hydrolysis. Organisms capable of hydrolyzing esculin react with ferric ions in the medium to form dark brown or black colonies, while Streptococcus agalactiae typically remains blue or colorless.
